Single-cycle kinetic evaluation

The multi-cycle kinetic (MCK) method is the most common method for kinetic evaluation. However, it is not easy to find the appropriate conditions for MCK analysis. In the case of the regeneration process, the analyte must be completely removed while not damaging the ligand.
If it is difficult to optimize the conditions for MCK analysis, single-cycle kinetic (SCK) analysis can be used. SCK analysis is performed by injecting analyte from the lowest concentration and gradually increasing the concentration within one cycle without the regeneration process.
We explained the characteristics of the MCK/SCK analysis method and investigated whether there were differences in binding characteristics.

As can be seen in the table above, the association rate constant (ka), dissociation rate constant (kd), and binding affinity (KD) all do not show significant differences depending on the analysis method. So, in which cases are each of the two analysis methods more suitable?

As mentioned above, when it is difficult to optimize the regeneration conditions, it is recommended to use the SCK analysis method. In order to find the appropriate regeneration conditions, you need to test various solutions with properties such as acid, base, ion, and hydrophobicity. In addition, in order to prevent damage to the ligand, you need to start with a solution with weak properties and gradually increase the strength while testing. For example, when using an acidic solution as a regeneration solution, you need to start with a high pH and gradually lower it to find the conditions for regeneration without damaging the ligand. Therefore, optimizing the regeneration conditions may require many tests, and even after this process, you may not be able to find the appropriate regeneration conditions. In this case, we recommend using the SCK analysis method that does not require a regeneration process. In addition, in SCK analysis, the dissociation of analytes of multiple concentrations in front is observed briefly, and the dissociation of the analyte of the highest concentration at the end is observed for a long time to evaluate the dissociation tendency. This helps reduce the total kinetic analysis time.
However, depending on the characteristics of the interaction, it may be difficult to evaluate the binding with only SCK, which has less information than MCK. In particular, the MCK method is suitable when the binding is complex. If one of the curves of multiple concentration analytes is bad, MCK has the advantage of being able to evaluate by removing the bad curve from the curve overlay.
